Samuel Rapoport was the son of Daniel and Manya (Weinbaum) Rapoport. He also had a sister, Betty (Rapoport) Rubinstein.
In 1933 he studied in Vienna, Austria, receiving his doctorate in medicine and chemistry. Starting in 1938, he was a pediatrician in Cincinnati, Ohio. He married Ingeborg Syllm, daughter of Paul and Rosa Maria (Fiebes) Syllm on August 15, 1946 in Cincinnati, Ohio and had 4 children together. In 1950, he and his famliy fled to East Germany from McCarthyite persecution in the United States. He was the director of the biochemistry institute for 30 years at Charité University Hospital in Berlin.
