Jacques de Savoie was the son of Philippe, Duc de Nemours, and his wife, Charlotte, Mademoiselle de Longueville, daughter of Louis I d'Orléans, Duc de Longueville. His father was a younger son of Philip II, Duke of Savoy, and his second wife Claudine de Brosse. He was married on April 29, 1566, to Anna d'Este, daughter of Ercole II, Duke of Ferrara and his wife, Renée of France, and the widow of François de Lorraine, Duc de Guise. They had three children: Charles Emmanuel, who died childless; Marguerite, who died in early childhood; and Henri, who succeeded his elder brother as the duc de Nemours.
