Edward was the son of Rev. Samuel Thompson & Sarah Shepard. He was born in Braintree, Suffolk county, Massachusetts Bay Colony.

Edward was a schoolmaster at Newburyport, Massachusetts. He graduated at Harvard College in 1684. He instructed at a school for several years at Newbury.

Edward married Sarah Webster in 1687 at Newbury, Essex county, Massachusetts Bay Colony. They had children:

Edward - 14 May 1695
William - 26 April 1697
John - 17 September 1699
Joseph - about 1704

He was ordained on October 14, 1696, when he was 31 years of age, and he became a minister at Marshfield, Plymouth county, Province of Massachusetts Bay.
