John Wesley Scarborough was born in Texas, the son of John Charles Henry Scarborough and his first wife, whose name is unknown. John C. H. Scarborough's family lived in Leon County Texas in the 1870s, but he remarried in 1876 in Freestone County Texas. Between 1880 and 1885, John C. H. Scarborough moved his family from Texas to Union Parish, where his brother, George Noah Scarborough, lived. They settled near what later became Spencer.

John Wesley Scarborough married on 20 July 1897 in Union Parish to Nancy Ammaline Guinn, daughter of Edward Wade Guinn and Josephine Samantha Kolb. John Wesley died between about 1900 and 1905 and is believed to have been buried in the Liberty Cemetery, where his father and siblings were buried.
