Mustered into service at Harrisburg on January 25, 1864, and mustered out on July 13, 1865.

The 3-year service men of the 20th Cavalry mustered in at Harrisburg and Philadelphia in January and February 1864. They reported to Gen. Sigel in the Shenandoah Valley and were engaged at New Market in May and at Staunton, New Glasgow, Lynchburg, and Salem in June. The regiment lost 14 at Snicker's Gap in July and 108 were killed, wounded, and missing a few days later in an attack on Early's column at Ashby's Gap. Near the close of February 1865, the 20th rode with Sheridan in the cavalry raid towards Lynchburg. A month later they joined the army in front of Petersburg, then starting its final campaign, with the cavalry in advance. Led by Col. Middleton, the 20th "was almost incessantly engaged and constantly in motion" in the fighting that led to final surrender on the 9th. On June 17, 1865, it was consolidated with the 2nd Regiment, Pennsylvania Cavalry, to form the 1st Pennsylvania Cavalry, and was mustered out on July 13, 1865, at Cloud's Mills, Virginia.
