Husband of Elizabeth Clay. John Clayton is the last ancestor of this Clayton line with documented proof. After extensive research at the Library of Virginia, no documentation was found to connect him to a father. Documentation was lost or destroyed. There are parallel Clayton lines, but none connect to John Clayton with documentation.

John Clayton apparently came to Isle of Wight County from Gloucester County. Entries in the Register of Abingdon Parish, Gloucester County record the "baptism of John Clayton, son of John and Elizabeth Clayton on February 2, 1717/1718;" the baptism of "Mary, daughter of John Clayton" on December 6, 1719; the birth of "Susanna, daughter of John Clayton" on February 28, 1722/1723; and the baptism of "Rebekah, daughter of John and Elizabeth Clayton" on June 5, 1726.

John's will was recorded in Isle of Wight County, Virginia Will Book 2, Page 191 in 1756, and proven on 4 Jan 1759, Isle of Wight, VA .

He named the following children, grandchildren, and sons-in-law in the order as written in his will:
