Reuben Cantrell was the son of Abraham, Jr. and Sarah Kilgore Cantrell. Reuben's father died shortly after his birth and when his mother remarried he and his brother Isaac were raised by his grandfather Abraham Cantrell Sr. and second wife Lucy Burke Anderson Cantrell (widow of Reuben Anderson and daughter of John and Mary Stevens Burke) in Pike-Letcher County, KY area.

Ruben married first to Nancy Francisco and second, at age 80, to Mrs. Cornelia White, a widow.

In 1860 Ruben and Nancy were recorded in Pike Co, KY and his real estate was valued at $160 and his personal property at $50. In 1870 they were recorded in the 9th District P. O. Robinson Creek and his occupation was listed as Baptist minister. In 1880 they were recorded in the Marrowbone Voting District and he was listed as a farmer.

In 1900 Ruben was not listed but Nancy was living in the home of son-in-law, John Bartley.
