Isabella de Clare was the eldest daughter of Gilbert "the red" de Clare, 6th Earl of Gloucester, 7th Earl of Hertford, 9th Lord of Clare, 3rd Lord of Glamorgan (1243 - 1295) and Alice "of Angoulême" de Lusignan (1236 - 1290). She married twice. Her first marriage was to Guy de Beauchamp, 10th Earl of Warwick (1272 - 1315) on 11 May 1279 at Warwick Castle, Warwickshire, England but was later annulled and found to be invalid. They had no children.

Her second marriage was to Maurice "the magnanimous" de Berkeley, 2nd Baron Berkeley, Warden of Gloucestershire (1271 - 1326) on 06 May 1314 at Berkeley Castle, Gloucestershire, England. Her marriage include the Lordship of Shipton and the Lordship of Buford. Lord de Berkeley died ten years later in prison at Wallingford Castle and they had no known children. She died childless.
