James Edward Duke was born on December 12, 1871 in Buffalo Valley, Putnam County, Tennessee, the son of William Wade Duke II (1846–1928) and Virginia Elisabeth "Betty" (Christian) Duke (1848–1925).

He was married in 1892 to Ida Anne Nichols (1876–1961), the daughter of Reverend Brice Byrne "B B" Nichols (1842–1923) and Margaret Coates (Allen) Nichols (1845–1926).

A self-employed automobile body builder, James was 61 years old and married when he injured his head in an automobile accident on October 4, 1933 in South Pittsburg, Marion County, Tennessee, and he died October 11th , and burial was two days later in Rocky Springs Cemetery, Bridgeport, Alabama.
