On August 18, 1825 in kentucky, James married Nancy Murphy. Nancy died in 1848 during James's 1st attempt to leave Missouri for California staying in Texas for awhile. James returned to Missouri where he married Martha E. Carter April 29, 1849 in Ray County, MO. He had two children with Martha Carter, Presley Bascom Barker and Sarah Barker.

James came to Oregon from Missouri by way of Texas to join his sons, James M. and Joshua. James, a Methodist preacher, kept a diary of his trip from Texas to Oregon in 1856-58 which has been published by the Lompoc County (Calif) Historical Society.
James's brother Henry W. Barker died on the plains in 1852 of cholera
