William Cox came to Virginia at the age of 12 on the 49-foot merchant vessel Godspeed, which arrived on 10 June 1610, under the headright system, sponsored by Matthew Edlow, Sr.

He was listed in the household of Thomas Boulding in Elizabeth City [County] in 1625.

A planter (farmer), he signed a ten-year lease for 100 acres of land in Elizabeth City [County] on 20 September 1628.

He received 150 acres of land near the James River above Arrowhattocks in Henrico County on 29 November 1636 for transporting three colonists to Virginia. He patented an additional 150 acres above Arrowhattocks on 29 October 1637.

William Cox bought 250 acres at the mouth of Falling Creek along the James River in Henrico County (present-day Chesterfield County) on 1 September 1642 in partnership with Isaac Hutchins.

He died before 14 December 1656, when Peter Lee patented 126 acres in Henrico County adjacent "land belonging to ye orphans of William Cox" (Henrico County, Virginia Patent Book 4, page 44).
