Wayne was the son of William and Elizabeth R. Hurst Harrell. He married Eliza Cunningham on 26 Jun 1847 in Claiborne County, Tennessee. They had been married less than one and one-half years when Eliza died, leaving him with a baby girl, Elizabeth Harrell. He married Malinda J. Bartlett on 05 Jan 1849 in Claiborne County, Tennessee, less than two months after Eliza's death. By 1852 the family had moved to Knox County, Tennessee, and by 1854 they had moved to Indiana. There were in Urbana, Dallas County, Missouri, by 24 Aug 1860 when the Federal census was taken. In Urbana, Wayne operated a water-powered mill.

Wayne served in the United States Army during the Civil War. He enlisted on 01 Nov 1863 and served as a sergeant in Company D., 15th Regiment, Missouri Cavalry Volunteers. He was discharged on 01 Jul 1865.

In 1874 they moved to a farm southwest of Urbana and he built a house there; they were still living there when he died. He told his family that he wanted to be buried in the courner of Mr. Bullock's field. He was the first person to be buried in what is now Mission Ridge Cemetery, south of Urbana, Dallas County, Missouri.

Wayne and Eliza had the one child, Elizabeth, born in 1848. Wayne and Matilda had ten children: Mary J., William Frank, Nancy Jane, Douglas, Daniel A., Dora Isabell, Lavesta Cornelia, Charles Andrew, Rosetta, and Josilyn Harrell.
