John Keyser was the eighth of ten children born to Carl/Charles Sebastian Keyser (1726/27-1778) and his wife, whose identity is disputed but may have been Mary Shelly. John's father was a native of Möckmühl, Württemberg, Germany and arrived in Philadelphia in 1749 Before John's birth his family moved from Lancaster, PA into Virginia's Shenandoah Valley. The family farm was situated in what was originally Frederick Co, but is now Page Co. near present-day Luray.

On August 30, 1791 John married Catherine Rinehart, known as Caty, a neighbor whose grandfather was born in Switzerland. John and Caty settled on a farm near Alma in Page Co., where they raised three children: Elizabeth Ann (b.1792), Alexander (b. 1794), and Christopher (b. 1799). The family also owned several enslaved people. Caty's will, dated 1854, names Keziah and her daughter Sara Virginia, a boy Henry, and an child Mary. John Keyser died at the age of 80, reportedly after suffering many years with rheumatism. The location of his grave site is unknown.
