Margaret was born in Bartholomew County, Indiana to John Henry and Frances Edna Hamner Blacketter. She married Henry Jasper Hamner on July 29, 1849 in Bartholomew County, Indiana. Margaret and Henry moved to Pike County, Illinois in the early 1850's. They farmed 160 acres in Hardin Township.

Margaret and Henry had 11 children: Martha Jane, Francis Macon, Sarah Rebecca, George W., Henry Jasper, Frances T., Rosa Elnora, Thomas Wilson, Joshua Emery, Rachel M., and Lucinda C.

After Henry Jasper died at the age of 46, she moved in with her son Thomas Wilson Hamner in Nebo.
