Son of Ralph de Neville and Joan de Beaufort. Grandson of Sir John de Neville and Maud de Percy, Sir Hugh Stafford and Phillipe de Beauchamp.

Robert was appointed Canon of Auckland and prebendary of Eldon in 1413, Canon of York and prebendary of Grindall in 1414, Canon of Howden Yorkshire in 1416, Rector of Spoffoth in 1417, Canon of Beverly in 1419, Provost of Beverly 1422, Canon of Lincoln 1424. He served as an envoy to Pope Martin V at the council of Constance 1418, made Bishop of Salisbury by papal provision at the request of his uncle, Cardinal Beaufort in 1427. He was made Bishop of Durham in 1438 on the recommendation of King Henry VI. He was formally installed into that office on 11 April 1441 in the presence of his brothers and a large assembly of nobles. Robert created the new offices of chamberlain, vice-chamberlain, master of the horse and armourer, apparently for the benefit of his relatives.

Robert de Neville, Bishop of Durham, was buried in the south aisle of the nave of Durham Cathedral.
