Alexander Humphrey was born in Connecticut, but migrated along with many others to the Catskills region of New York shortly after the Revolutionary War. He was a road builder who worked with several of his brothers. His father was Captain Frederick Humphrey, and his mother was Ruth Tuller Humphrey of Connecticut.

Alexander married Roxana "Roxey" Brown, daughter of Captain John Brown and Hannah Owen Brown. According to the Humphreys Family In America text, Alexander and Roxey migrated initially to Durham, Greene, New York, then moved a few miles further to Manorkill, Schoharie, New York, where they owned a farm and ran a hotel.
