Enlisted in Alamance County, North Carolina at age 22 with Company F on June 10, 1861 for the war. Mustered in as a Private and appointed Quartermaster Sergeant September 14, 1861 and transferred to the Field and Staff. On November 29, 1861 Mebane Cornelius Mebane was promoted to Sergeant Major.

He was wounded at the Battle of South Mountain, Maryland. He soon was promoted to Adjutant with the rank of 1st Lieutenant on September 16, 1862. He received a second wound at the battle of Rappahannock Station, Virginia on November 7, 1863 and carried as absent wounded through December 1864.
