Nathan was the youngest known son of Rev. William May and Elizabeth "Betsey" Browning May. He was born and raised in Lowndes County, Alabama. His father died in 1825 when Nathan was four years old. His mother remarried to John Sessions who died in 1835 when Nathan was about 14 years old. By 1850, his older brother, Asa Thomas May, had died and Nathan eventually became the guardian of Asa's children.

Nathan enlisted in as a Private in Company E (later Company M), 6th Alabama Infantry and was killed at Cold Harbor. He left a will that was probated in Lowndes County, Alabama. He named his wife, Dorothy Susan. He married her after the 1860 census as he was still single, living in Lowndes County, Alabama then. If he had no heirs by Dorothy, then half of the estate went to her and the other half to his brothers John Browning May, William Smith May, David Francis May, and his niece, Nancy Cornelia Haralson. Nancy was the daughter of Nathan's late sister, Nancy Ann May Haralson, and her husband, Elijah "Eli" Haralson.
