John Hopkins was married to Amner Sexton in 1838 or 1839. She died on 29 Mar 1878 and he then married Nancy M. Welch in 1879. He and Amner had thirteen children; Mellinda, Benjamin, John Calvin, William A., Hiram Silas, James Taylor, Nathan Jackson, Margaret Jane, Nancy Elen, Harriet Frisby, Mary Telitha, Lydia Ann, and Samuel H. Hopkins. He and Nancy Welch had four; Helen, Porter J., Rebecca E., Lillie Pearl Hopkins.

He was ordained as a minister, or revivalist, in October of 1859. He conducted many revival meetings in Kentucky and in Tennessee. He was pastor of Black Creek church in Scott County, Tn for several years.Baptist Minister
