Eli was the son of William H. "Buck" Landreth and Majorie Cox. He married Susannah Faddis of Grayson Co., VA, on 17 November, 1854.

Eli Landreth resided Washington County, VA. He enlisted in Co. A, Virginia, 51st Infantry Regiment on July 16, 1861 as a Private in Abingdon, Virginia. He was 5'9", had sallow complexion, brown eyes, and dark hair. (Source: "The Virginia Regimental Histories Series")
