Duryea was the oldest son of Hollywood Actor, Dan Duryea, and Helen Bryan. He had many occupations during his life, having been a writer, director, documentary filmmaker, boat guide, and naturalist.

Before trying his hand at acting, he majored in math and physics at Amherst College in Massachusetts. He appeared in plays in Houston and New York before moving back to Hollywood, where he acted in two films and an episode of "The Daniel Boone Show" with his father. Though Peter Duryea appeared in six films and thirty television shows, the pace of the hectic Hollywood lifestyle was too much for him. He moved to Canada in 1973.

Duryea's most notable role was that of Lt. Jose Tyler, navigator of the USS Enterprise, in the pilot episode of "Star Trek".

He settled in Gray Creek, British Columbia, and became involved with various community projects that would carry on for the rest of his life. He founded a children's theater group called the "Kootenay Lake Players" and served as a volunteer board member of the "Kootenay Lake East Shore Eldercare Co-op".

Duryea's most lasting achievement was the founding and building of the "Guiding Hands Recreation Society's Tipi Camp and Nature Retreat." He was very well respected by the local community, as stated in the local paper: "It's a measure of Peter Duryea's worth that appearing in one of the most popular shows in television history wasn't his most noteworthy accomplishment. To most of the world, Duryea was best known for a bit role in the pilot episode of the original Star Trek series. But on Kootenay Lake, he was a revered East Shore elder and environmentalist who fought against clearcut logging and started a now-thriving nature retreat."

Duryea died at home after a long illness. Private Family Service and Burial on Thursday, 28 March 2013, at the Gray Creek Cemetery.
