Knox Sixteen Church was built in 1845 in the Village of Sixteen on a 1/4 acre of donated land. The adjacent lot was purchased for a cemetery.

The once thriving community disappeared during the 1870s as Oakville was being developed but the local farmers continued to maintain their church. It was part of a two-point charge with Knox Presbyterian Church for 50 years to 1883, thus the name Knox Sixteen. The building was enlarged and bricked over in 1899, the interior redecorated in 1925, electric lights installed in 1943 and in 1994, a basement was added beneath the original structure.

The church is still in use today.
