Karoline Kolmer was born in North Carolina, on 31 December, 1838. She was the third child of Michael Kolmer and his wife Josephine Wagner who had emigrated from Endingen, Baden, Germany, in 1833.

Michael, Josephine and family lived in Missouri in about 1841 and then traveled west in about 1845 to the Sacramento area. In about 1846, the family settled near Occidental and later relocated to a farm near Fort Ross at Timber Cove.
