Basil Prather was born to Aaron Allen Prather and wife Jane (a cousin) on 20 October 1742, in Prince George's County, Maryland. Paternal grandparents are documented as Thomas McKay Prather and Martha Sprigg. Maternal grandparents are believed to be Jonathan Prather and Elizabeth Biggers.

Basil grew up in a large family, with a twin named Baruch, and ten other siblings. Earlier church records for this family are found in All Hallows Parish, Anne Arundel County, Maryland. Land records indicate that the family moved to Frederick County, Maryland, by 1761, and later to Montgomery County, Maryland. A cemetery that holds many of Basil's family members in unmarked or unidentified graves is located on the property of the Prather homestead in Rockville, Montgomery County, Maryland. It is probable that his parents are buried there.

About 1765, Basil married Clorenda Robertson, and together they had twelve known children. They include: William (b.1766, m. Ary Gaither and Lettice McCarroll), Walter (b.1768, m. Martha "Patty" Jacobs), Thomas (b.1769, m. Rachel Gaither), Basil R. (b.1771, m. Rachel Holman), Samuel (m. Edieann Holman), Chlorenda (b.1774), Lloyd (b.1782, m. Nancy Redmon), Simon (b.1791, m. Elizabeth Simmons), John (m. Ann Campbell), Aaron, Asa and Susannah.

Basil was a Patriot, serving with the North Carolina militia (Salisbury District) during the Revolutionary War. In 1790, the family is found on records residing in Iredell County, North Carolina. In 1804, the family moved a final time, to Clark County, Indiana.
