Alfred Mitchell Barton was the son of Riley Barton (1826-1863) and Mary Ann Cantrell (1835-1862). Mary Ann was a direct descendant of William Cantrell, who came to Jamestown, Virginia, in 1608.

Alfred Mitchell Barton was orphaned after his family moved to Augusta, Arkansas; his grandfather, mother, and father died within just a few months of each other in 1862-1863.

Alfred Mitchell Barton studied medicine at Louisville College of Medicine in Kentucky.
