Henry was born Honore Dufresne. He was the son of Basile Dufresne and Marie Louise Benoit. His family migrated to the United States probably about 1851 when a group of French Canadians arrived in St. Anne, Kankakee County, Illinois.

Henry married Sophie Boisvert, daughter of Joseph Boisvert and Marie Archange Caille, on 9 Jan 1854 at St. Anne, Kankakee County, Illinois. Four sons were born to this marriage; Henry Dufrain Jr., Charles J. Dufran, Louis Jerry Dufrain and Edward Carry Dufrin. (The spelling of the name Dufrain was changed by one son to Dufrin and another son spelled it Dufran.)

Between 1870 and 1875 Henry and Sophie, along with their four sons and other French Canadian families, left Illinois and moved on to the Cloud County, Kansas area. They settled in Colfax Township and later Shirley Township.
In 1896, after his wife, Sophie was killed in a tornado, Henry moved to Hartington, Nebraska to live with his son, Henry Jr. He lived there until his death in 1916.
