John MacDonald was the son of John and Sarah (Dew) McDonald.

He married Mary Potter on July 3, 1854 in Perry Co., OH. They had at least six children:

- Sheldon St. Clair MacDonald (1855-1912)
- Chestora MacDonald Carr (1862-1948)
- Chester Cecil MacDonald (1865-1888)
- William Nial MacDonald (1868-1951)
- Alverda May MacDonald Strickland (1871-1963)
- Topsy MacDonald (1874-bef 1888) - she is the only sibling not mentioned in Chester's 1888 will
