Born in Scotland, he moved with his family to Canada when he was 14. He arrived in Coloma in the autumn of 1850, where he worked as a miner and baker, saving enough money to return to Canada for his family. When he got back to Coloma he bought the Sierra Nevada House, expanding the hotel he operated until 1865. His wife Catherine died in 1857, and two years later he married Delia Kneeland. After Delia's death he married his friend Martin Allhoff's widow, Louisa, adding on to Martin's vineyards and building Coloma's Vineyard House. He was elected El Dorado County Treasurer in 1867 and to the State Legislature in 1871-72.

Robert Chalmers also owned the land where the Coloma Pioneer Cemetery is located.
