Stephen was the son of John M. Vanderpool and Sarah Elizabeth Cummings.

He married Margaret Reynolds (1835-1865) in 1852, Rockcastle County, KY, and their children were Cynthia and Martha J. Vanderpool.

He married Mary Catherine Potts (1847-1926) on 4 Jul 1867 in Lincoln County, KY, and their children were Sallie and Lena. After Stephen's death, Mary married J. M. Taylor on 16 Jun 1910 in Groesbeck, TX.
