Margaret was the daughter of Dr M. Q. Ashby of Kentucky and grew up in Mount Sterling, Kentucky. In 1839 she married Samuel Hughes Woodson, a young lawyer, and they lived briefly in Nicholasville, Kentucky. Upon moving to Independence, Missouri in 1840 they lived in a log cabin.

Her husband was a member of the law firm of Woodson, Chrisman & Comingo, later elected to Congress, and then to the circuit court of Jackson County.

Margaret and Samuel Woodson had 11 children. At the time of her death at age 92, she was survived by five children: Mrs. Nellie Reese of Independence; Samuel H Woodson, Jr., an attorney and former mayor of Independence; Mrs. Henry Harper of Kansas City; Mrs. A. F. Sawyer of Independence; and William H. Woodson, a lawyer in Liberty, Missouri.
