Margaret was the daughter of Dr M. Q. Ashby of Kentucky and grew up in Mount Sterling, Kentucky. In 1839 she married Samuel Hughes Woodson, a young lawyer, and they lived briefly in Nicholasville, Kentucky. Upon moving to Independence, Missouri in 1840 they lived in a log cabin.
Her husband was a member of the law firm of Woodson, Chrisman & Comingo, later elected to Congress, and then to the circuit court of Jackson County.

At the time of her death at age 92, she was survived by five children: Mrs. Nellie Reese of Independence; Samuel H Woodson, Jr., an attorney and former mayor of Independence; Mrs. Henry Harper of Kansas City; Mrs. A. F. Sawyer of Independence; and William H. Woodson, a lawyer in Liberty, Missouri.
