She was the daughter of Louis-Emmanuel de Valois, Duc d'Angoulême, and his wife, Marie Henriette de La Guiche, Dame de Chaumont (died 1682), daughter of Philibert de La Guiche. Her father was the son of Charles de Valois, Duc d'd'Angoulême (the illegitimate son of King Charles IX and Marie Touchet) and Charlotte de Montmorency.

Marie Françoise de Valois was married on November 3, 1649 at Toulon to Louis de Lorraine, Duc de Joyeuse. She succeeded her father in 1653 and became the 3rd Duchesse d'Angoulême. She was confined to the abbey of Essey in the Côte-d'Or department in eastern France because of mental instability.
