She is the daughter of Joshua and Elizabeth (Elmore) Stapleton. Military records show that Joshua was a private in the Indian War of 1811. Gibson County Historian Elia W. Peattie notes that he was "a hero in the Battle of Tippecanoe." Fellow historian Gil R. Stormont says the family settled five miles east of Princeton in 1816.

Joshua farmed 160 acres of land on which stood the Lawrence Schoolhouse. This is "McConnell's Corner" northwest of Francisco, IN. He gave 80 acres to Louisa upon her marriage to Duston Mills. They were neighbors across CR 450E/Fairview Rd with the Mills, who settled the area around the same time (most likely 1811). Both families were pioneers of that area prior to Indiana statehood. The Stapletons were from Robertson County, TN. The Mills came from Kennebec County, ME.

She and Duston married 16 December 1827 in Gibson County, IN. She gave birth to 12 children, including three sets of twins, the firstborn son died in childbirth (Zyasa). They farmed in Section 11 of what became Center Township in 1880. She outlived Duston by 7 years and 2 months. She took care of their mentally disabled daughter, Elvira, until Louisa's death in 1882.

Assuming her middle name may have been Elizabeth, since she's often referred to as "Eliza."
