Gladys Humphreys was the oldest child of Nathan Syrus and Margaret Delilah Fisher Kilgore. When she was 16, she won a piano in a contest sponsored by a grocery store in Munday, Texas. She began lessons with the piano teacher at school, who inspired her to become a teacher.

She began teaching piano and voice in 1919, and continued until her retirement in 1981 at the age of 85.

In 1927 she married William Robert Humphreys at Hereford, Deaf Smith Co., TX. They had three children: Zoe Evelyn (marr. Roy Maxell Smith), Anna Ruth (marr. Ward Byrd Benson Jr.), and William Howard (marr. Anna Jeanne Troxell).
