John was the son of John Wesley HUFFORD and Harriet GOODING. His siblings were Charles W. (1868-1949), Elizabeth (b. 1868, m. NUZUM), Minnie (b. about 1871), and Ettie (b. about 1873). His maternal half-siblings from his mother's first marriage were William James VAUGHN (b. 1857), Alice Vaughn (b. about 1858), Hattie Vaughn (b. about 1862), and Alonzo Vaughn (b. about 1864).

John was married twice. His first wife is unknown; a daughter was born to that marriage, Ethel (b. about 1901). On January 19, 1907, in Columbia Co., Pennsylvania, at the Wesley United Methodist Church, John married Bessie C. LOFTUS. Born to his second marriage were L. Charles (b. about 1910), Bessie (b. about 1910), Bowman John (1911-1989), Audrey Mae (b. about 1914), Howard H. (1915-1991), and Woodrow (1917-2000). In 1920, John was working as a structural worker at an iron mill. By 1930, he was working as a lock tender.

John's paternal uncles (brothers of his father) were William Hamilton Hufford (1847-1906) and Joseph D. Hufford (1852-1925, m. Adelia SWORDS). John's great-uncles (brothers of his grandfather George) were John Hufford (b. 1823, m. Eliza JELLISON) and Henry Hufford (b. 1825, m. Sarah Ann WOLFORD).
