Son of Francis Cokayne and Dorothy Marrow. Husband of Dorothy Ferres, the daughter of Sir Humphrey Ferrers. They had three sons and seven daughters.

Sir Thomas was invested as a knight in 1544. He was one of the founders of the Queen Elizabeth's Grammar School in 1592. He also acted as a guard for Mary Queen of Scots when she was sent from Wingfield Manor to Tutbury Castle.

His tomb, located outside the Boothby Chapel on the north wall of the north transept is the most magnificent and dominant tomb, yet his will dictated that he wanted 'no vain pomp nor foolish ceremonies' for his funeral and that 'instead of a tomb...set in the wall by my father's tomb in my choir at Ashborne church some small monument of brass'. The names of his children are also listed.
