Joanna was the second child to John Bryce Cook and Catherine Kellog Keeler Cook. Family lore says that Joanna had scarlet fever as a child, which left her partially deaf. She was very small in stature, which was also attributed to her childhood illness.

Joanna was a very religious woman, who never married. She lived with her family for her entire life, and passed away in her home.

She was buried at Rosedale Cemetery, in the family plot, along with her parents, brother Edward Thayer Cook, sister-in-law Mary Ellen Woods Cook Beall, and nieces and nephews.
