Nelson was the son of Robert and Susan Woodbury Berkley. His father was a native of Scotland. His grandfather, also Robert, was born 15 April 1756 in Scotland and married Rhoda Way who was born in Sharon Connecticut 21 August 1759. They had children: Rhoda(17 March 1780), William(5 January 1783), Anna(8 January 1785), Elizabeth(10 October 1788), Irena(10 July 1790), Cyrus(5 July 1792), Hannah(1 November 1795), Robert(1 September 1798), and Sarah(19 September 1801).

Robert and Susan were married 7 September 1820 in New Hampshire. They had seven children: Britain(9 August 1821), Arthur(25 January 1823), Ladora(28 February 1824), Oscar(28 January 1830), Ladora II(24 January 1836), and Thirzah (24 September 1839). The father was a shoemaker, operated a tannery, a glass factory, and a sawmill and gristmill. He died in Redfern, New York on 18 October 1853. His first wife had died many years previously and he was living with his second at the time of his death.

Nelson came to Ohio in August 1850 after being employed by his father making boxes for the glass factory and in the saw and grist mills. He married Mary Ann Bigelow on 1 November 1849 in New York and traveled the entire distance to Ohio with two horses, a light wagon, and a buggy. He settled in Medina County for about three years where he was in charge of different sawmills and engaged in breaking colts and trading in horses. He returned to New York with his wife and children because of his health and lived in Clinton County. After two years, he went to Neenah, Wisconsin where he was first employed as a carpenter. In the fall, he returned to New York and brought his family back to Wood County.

The children born of the first marriage were Van Rensselaer born in Medina County 7 September 1850 and died 23 February 1851, Helen Virginia born 25 March 1852 and married George Sykes, Ladora Imogene born in Wood County on 5 April 1855 and wife of Samuel Wood, Mary Frances born in Saranac, New York, 20 May 1857 and wife of John Wolf; Robert Nelson born in Clinton County, New York 9 March 1860 and died 18 June 1864; Susan Ellen born 10 October 1862 in Wood County and married Jacob Hock; Ernest Elmer born in Wood County 9 February 1865; and Elizabeth Alta born 19 October 1867 and married Samuel Smith.

After Mary Ann's death in 1883, he married Mary Ann Landis on 30 December 1885 in Wood County.
