Hon. John Spencer was born in Althorpe, Northamptonshire, England and was the son of Sir Charles Spencer, 3rd Earl of Sunderland and Lady Anne Churchill. He married Lady Georgiana Caroline Carteret, daughter of John Carteret, 2nd Earl of Granville and Frances Worsley, on 14 February 1733/34. Hon. John Spencer held the office of Member of Parliament (M.P.) for Woodstock.

The couple had issue: John Spencer, 1st Earl Spencer and Diana.

He passed away in Wimbledon, Surrey, England. He was buried on 30 Jun 1746 in , Brington, Surrey, England.
