He was the son of Enos McHenry Hooper and Matilda Burrell. Some earlier census records have him as Henry perhaps to differentiate him from his father who was also Enos McHenry Hooper. He married Mary Adeline Ammons in Cherokee, North Carolina, USA. She was the daughter of Rev. John Peake and Narcissa Caroline (Rogers) Ammon.

Enos Hooper, resident of Towns County, Georgia, enlisted 27 Sep 1862 for the Confederacy serving the state of Georgia. He enlisted as a Private on 27 September 1862 in Company G, Smith's Legion Infantry Regiment Georgia. He later transferred into Company G, 65th Infantry Regiment Georgia on 15 Mar 1863.
Transferred out of Company G, Smith's Legion Infantry Regiment Georgia on 15 Mar 1863.
