William Darby was born in 1731 to William Darby II and Mary Unknown Darby in Scotch Plains, New Jersey Colony. His father was a miller and land owner and among the founders of the Scotch Plains Baptist Church to which he donated land for their meeting hall, parsonage and cemetery.

William Darby married Patience Frazee in 1752, they had two daughters, Mary and Hannah. After Patience's death, he married a second time to Sarah Sayre Owens, widow, and had a son named Ezra. Ezra went on to become a US Congressman during the 9th and 10th Congress.

On Sept. 1, 1775, William Darby was a Freeholder of Elizabethtown, Union County, New Jersey. On Dec 29, 1780, along with David Morris, he was a executor of the estate of Recompense Stanbury. On Nov 28, 1789, he was named a member of the Common Council of the Borough of Elizabeth, which at that time included all of what is now Union County. William Darby died on November 10, 1793.
