Elizur was baptized at St. Lawrence Jewry in London, England on 4 May 1617, and was 20 years old when he arrived in the Massachusetts Bay Colony in 1637 or 1638. He was the son of Edward Holyoke Jr. and Prudence Stockton. He was a farmer and land owner. He married Mary Pynchon, daughter of William Pynchon, in 1640 in Springfield. The couple had eight children: John in 1641 (died in infancy), John in 1642, Hannah in 1644, a stillborn daughter in 1646, Samuel in 1647, Edward in 1649, Elizur Jr. in 1651 and Mary in 1656.

After his wife's death, Elizur married the widowed Editha Stebbings Maynard Day in 1658 in Springfield. He became the second largest landholder in Springfield, which had been founded by his father-in-law. He held the offices of commissioner, surveyor, deputy to the general court, associate county judge, and selectman. Mount Holyoke was named after him, and indirectly, Mount Holyoke College and the city of Holyoke (which were named after the mountain).
