Benjamin was the fifth of the eleven known children of John and Susannah (Countryman) Ambrose. His birthplace is unknown but was probably Somerset Co. PA since he was baptized in the Berlin Lutheran Church there. Some time after that, Benjamin's parents moved to a farm near Ligonier in Westmoreland Co. PA.

Benjamin married Ann Shearer. They moved to Armstrong County in time to be listed on the 1820 census for Sugarcreek Township.

Benjamin and Ann had the following children:
- Mary B. (Ambrose) Foster (1818-1913)
- John Ambrose (1819-1909)
- Eliza (Ambrose) Mateer (1823-1903)
- David Ambrose (abt 1825-1852)
- Susannah (Ambrose) Leard (1827-1849)
- Frank Ambrose (1830-1906)
- Joseph Ritner Ambrose (1839-1910)

After Ann's death in 1842, Benjamin married Peggy Campbell, who survived him.
