Rodolfo Amedeo Lanciani was the son of Pietro Lanciani and Lucia Galardi de Jagellis.

Rodolfo had three brothers, Filippo, Camillo, Cesare and a sister, Carlotta.

He first married Mary Ellen Rhodes in Paris, France, in 1875. Mary Ellen Rhodes was the daughter of Pelig Aborn Rhodes and Elizabeth Avery of Providence, Rhode Island, USA. Mary Ellen was born in 1843 and died on 14 February 1914, in Rome.

Mary and Rudolfo had one daughter, Marcella, and three grandchildren.

Lanciani remarried on 9 October 1920 to the widow of Prince Marco Antonio Colonna, Princess Teresa Caracciolo in Rome. Princess Teresa Caracciolo Colonna Lanciani was born on 5 November 1855 at Naples and died on 28 June 1935 at Rome. She was the daughter of Luigi Caracciolo, Duke of San Teodoro and St. Arpino and Augusta Selina "Leila" Lock.

The children of Princess Teresa Caracciolo Colonna and Prince Marco Antonio Colonna were:
1. Isabella Colonna (1879 -1957)
2. Victoria Colonna (1880-1935)
3. Stefano Colonna (1884-1885)

Lanciani held degrees in philosophy from the College of Rome and a degree in philosophy and mathematics from the University of Rome. He was a Professor of Surveying at the University of Rome, from 29 November 1894 until his retirement in 1928. He served on the City Council of Rome from 1914-1920 and he was the Deputy Mayor of Rome.

Rudolfo Lanciani was an archaeologist and art historian. He was known for his studies of the topography and monuments of the city of Rome.

He was prominent in excavation work at the site of ancient Rome and occupied the chair of science at the University of Rome. He also lectured at Harvard College, Cambridge, Massachusetts, USA.

Rudolfo Lanciani authored many books in English, including;

"Esegui ancor giovane gli scavi di Porto per i Torlonia e dal 1871 fu segretario della Commissione Archeologica Comunale, compiendo un serie di scavi a gara con la convulsa espansione edilizia della città , seguendo, da un cantiere all'altro, il venire alla luce e l'immediato scomparier di interi quartieri dell'antica Roma tra il 1871 e il 1890, di ogni scoperta dette via via comunicazione nell' organ municipale, cosi che è stato possiblil avere ampia conoscenza delle strutture antiche della città ormai definivamente ricoperte Docente di Topografia romana all' università La Sapienza, ha rielaborato la topografia dell' antica Roma nella Forma Ubis Romae, pubblicata tra il 1891 e il 1901, e ha scritto una Storia degli scavi di Roma, in quattro volumi editi tra il 1904 e il 1910 e relativi al perido dal 1000 al 1605, mentre sono stati pubblicati postumi quelli relatvi al periodo dal 1605 al 1879."
