John Barrett was born on June 12, 1708, the eighth child of Samuel and Sarah (Manning) Barrett. Most likely he was born in Boston, where his father was a merchant, who lived on Hanover Street.

John also became a Boston merchant, and by the late 1750's he was wealthy enough to commission John Singleton Copley to paint two handsome three-quarter length portraits of himself and his wife Sarah . He was "one of the Committee of Six, appointed by the merchants of Boston, in 1768, to resist the imposition by Parliament upon the Colonies of the onerous duties on sundry articles of commerce." According to his obituary in the Boston Gazette, "As a patriot, he was deeply interested in the freedom and happiness of his country, and sacrificed an independent fortune, the labour (sic) of his whole life, to promote their establishment."

He was for many years a deacon of the New North Church on Hanover Street , and for over thirty years he was one of the "overseers of the poor" .

He married Sarah Gerrish, daughter of Capt. John and Sarah (Hobbes) Gerrish, on June 3, 1731, with Dr. Joseph Sewall performing the ceremony. Together he and Sarah had eleven children.

He died on September 9, 1786 and was buried in Tomb No. 127 in the Granary Burial Ground in Boston.
