James Monroe Culpepper (1838-1901), son of Mariner (1793-1838) and Jane McCrary Culpepper (1798-1874), was born in Monroe County, Georgia, the youngest of nine children. His siblings included Bartley Anderson Culpepper (1818-1821), Pallitine Caroline Culpepper (1820-1821), Comfort Amanda Culpepper (1822-1868), Daniel Clark Culpepper (1825-1862), Benjamin Franklin Drayton Culpepper (1828-1839), Mariner Jackson Culpepper (1830-1872), Lucinda Council Culpepper (1832), and Frances Edna Culpepper (1834). After his father's death in 1838, his mother moved the family to Houston County, Georgia.

James Monroe Culpepper served in the 6th Georgia Infantry Regiment during the Civil War, enlisting in Atlanta in 1861 and serving until 1865. After the war he served as Senator from the 23rd Senatorial District and was a member of the Houston County Board of Commissioners and on the Fort Valley City Council.

He married Martha Kemp (ca. 1846-1870), the daughter of Joseph (1813-1860) and Catherine Elizabeth Kemp (1826-1906) in 1866. Catherine Elizabeth Kemp was the daughter of James T. Davis and Hannah Kinsey. After his first wife's death, he married Nancy Hannah Kemp (1857-1917) in 1876.
