Chamberlin was born at Brainerd Mission,TN. (at what is now Chattanooga, Tn).
Both Chamberlin's father and grandfather were missionaries to the Cherokees in their eastern homelands and assisted in the removal of the Cherokees to Indian Territory. Chamberlin married his cousin, Dolly (Dollie) Eunice Hoyt, in 1843, and the couple had several children. Hoyt was the granddaughter of former assistant chief of the Cherokee Nation George Lowery.
