Many researchers believe Samuel is the son of John Painter (Rev War Pensioner from NC). Although possible, I have not found any documents that support the theory.
He did marry Elizabeth Briance, possibly in North Carolina around 1795. By the late 1820's, they had moved to Montgomery County, Illinois. He eventually settled in Springfield, Missouri - possibly as early as 1831 - for sure by 1833.
Samuel and his wife Betsy were buried where the First Baptist Church now stands on South Street in Springfield and later transferred to unmarked graves in Hazelwood Cemetery.
They were the parents of at least six children; two were Elisha Painter and Jacob Painter
