The first of Alexander and Catherine Macomb's 10 children, he was born in what was then a colony of France in North America. On March 29, 1797, he married Christina Livingston, daughter of Ph. Philip Livingston, in NYC. That same year in NYC they sat for portraits by St Memin. In 1986,those were gifted to Clermont, the Livingston Manor, marking its 300th anniversary, by J.N. Macomb,V, the last in the direct line of J N. Macomb, I.

John was a merchant in NYC. While in transit on board the British packet "Princess Charlotte", he was killed defending her against a French privateer from the Isles of Scilly. His burial was at King Charles the Martyr Church in Falmouth, England and is marked by a tablet in the parish church commemorating his service and death.
