Nathan was the son of Jeffrey Preston and Jane Carter Burgess.

Nathan was married (firstly) on April 27, 1846, in Johnson County, Kentucky, to his second cousin Anjaline Preston.

He was married (secondly) on September 8, 1874, in Johnson County, Kentucky, to Jemima Ida (Osborne), widow of Andrew Jackson Price, his second cousin.

In June 1846 Eliza Hager filed Bastardy charges against Nathan Preston. The court dismissed it but it showed up again in the next session and it was again dismissed.

He was a merchant.

He enlisted December 12, 1862, as a Private in Company A, 39th Kentucky Infantry, Union Army, during the Civil War, and was discharged on September 16, 1865.
