In 1832, Holmes moved from Mercer County, Pennsylvania to Rush County, Indiana with his first wife, Mary Jane Wilson, where he purchased 220 acres. After his first wife's death in 1845, he married Ann Mariah Endecott on April 29, 1846. Holmes and Ann later moved to Howard County, Indiana.
